Moringa oleifera leaf extracts protect BMSC osteogenic induction following peroxidative damage by activating the PI3K/Akt/Foxo1 pathway

Abstract Objective We aimed to investigate the therapeutic effects of Moringa oleifera leaf extracts on osteogenic induction rat bone marrow mesenchymal stem cells (BMSCs) following peroxidative damage and explore underlying mechanisms. Methods Conditioned medium was used induce differentiation BMSCs, which were treated with H 2 O , extracts-containing serum, or phosphatidyl inositol-3 kinase (PI3K) inhibitor wortmannin, alone in combination. Cell viability measured using MTT assay. cycle assayed flow cytometry. Expression levels Akt, phosphorylated (p)Akt, Foxo1, cleaved caspase-3 analyzed western blot analysis. The mRNA osteogenesis-associated genes, including alkaline phosphatase (ALP), collagen І, osteopontin (OPN), Runx2, detected qRT-PCR. Reactive oxygen species (ROS) malondialdehyde (MDA) levels, as well superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), ALP activity commercially available kits. Osteogenic capability determined alizarin red staining. Results During reduced cell proliferation, inhibited osteogenesis, increased ROS MDA decreased SOD GSH-PX activity. significantly pAkt Foxo1 expression, BMSCs. Additional treatments partially reversed -induced changes. Wortmannin attenuated protein expression pAkt, caspase-3, genes. Conclusion ameliorate enhance BMSCs by activating PI3K/Akt/Foxo1 pathway.
